Abstract
The enzymatic activity of L-lactate: NAD+ oxide-reductase (LDH; EC. 1.1.1.27) isoenzymes in each of the embryonic development stages of Betta splendens was determined. Two hundred eggs were taken in each embryonic stage (fertilized oocyte, cleavage, blastula, gastrula, and neurula) from four sexually mature fish. Additionally, unfertilized oocytes were collected from two other females. Each sample (200 embryos or 200 unfertilized eggs) was homogenized and spun for 15 minutes at 10,000 rpm. The supernatant was run by electrophoresis in agarose gels, and total LDH activity was measured spectrophotometrically. The electrophoregrams were scanned at 540 nm in a densitometer to calculate the LDH isoenzyme-specific activity. During the embryonic period, LDH-3 and LDH-4 were characterized by presenting the most enzymatic activity. LDH-2 was expressed in all stages of gastrula and neurula but showed less activity.
