Abstract

In this work a method was established for measuring the activity of PAL in crude extracts of coffee leaves. The product of the PAL reaction trans-cinnamic acid, was quantitated by high performance liquid chromatography, after its extraction with chloroform. The presence of the enzyme in leaves of resistant (Timor Hybrid and Colombia) and susceptible varieties (caturra) to Hemileia vastatrix Berk & Br., was studied. According to the results, there is no significant difference in the basal activity of PAL among rust resistant materials. In the varieties Colombia and Timor Hybrid; the values were 10,6 ± 3,0 and 13,18 ± pKat/mg protein, respectively. These resistant materials showed basal values of activity greater than those found for the Caturra variety, (3,57 ± 0,6 pKat/mg protein).
